Phagocytic activity, oxidative burst activity, and production of reactive oxygen species (ROS) were quantified. Proteome microarray analysis was performed to identify changes in protein expression between control and test cultures. Surfaces were observed with scanning electron microscopy. Bacterial cultures showed that all test surfaces were similar to control in regard to cellular toxicity, proliferation, and lactate dehydrogenase release. Biological activity was similar for all materials, with some minor differences. Cultures of test surfaces and control surfaces in co-culture exhibited similar patterns of activation of phagocytosis, oxidative burst activity, and production of ROS. Proteome analysis identified at least 48 proteins that were altered in expression as a result of test material exposure. Several proteins were related to activation of immune response and inflammatory response. In conclusion, the metal-on-metal articulation surfaces tested were shown to exhibit a similar toxic effect on primary human macrophages, with the exception of cobalt.
